There really is no need to separate them. Mama hen will separate herself when the chicks are able to care for themselves. There is no set time for that as each hen is different, but you can expect the hen to stop being mom around six to eight weeks. From there the chicks can become part of the flock or be sold/given away/etc.

A chickens' life is ruled by mother nature. As was previously mentioned the hen will separate herself from her chicks in her own good time and the pecking order will takeover after that.

Why do you want to separate them? You can take the chicks away as soon as they hatch, let Mama raise them for you, any time in between, or leave them in the same flock for life. You have all kinds of options. To be able to suggest which you might want to take we need to know your circumstances and why you want to separate them. We all have different goals and circumstances.
