I have never had a sick chicken so I don't even know where to begin. I noticed about two days ago a couple of black spots on her comb, now it has taken over Her whole comb and one eye and ear it seems.

Help???

It may very well be a fowl pox infection.  It's a virus which will eventually run its course.  Antibiotics are not needed unless she develops a secondary bacterial infection.  It is spread by mosquitoes, and your other hens may be bitten and infected by them.  Do an on site search on 'fowl pox' for more information.
