Ok, I bought 6 blue laced red Wyandottes, only two have rose combs. I will also be getting some more Wyandottes next month
Anyway, what I'm wondering is how hard is it to diminish the single comb out of my flock? 3 of the 6 I currently have will be sold as 4 of them are roosters (2 of which are the rose combs). So I'll have a rooster and 2 hens from that group. Should I definitely keep one of the rose comb roosters ?

Rose combs are dominant. That means that if you have single combed birds, they don't carry the genes for rose combs, and will not produce rose-comb chicks. If you're determined to have a rose-comb population, I would get rid of all of the chickens with single combs, and probably all of the ones from that batch—it's highly likely that even your rosecomb roosters carry the single comb gene.
