From hatch they always looked like little girls from the start but now I figure is a better time to check!

Any ideas?

Now I have two adult serama hens: one has a large red comb and waddles, and the other hen has a small comb and no waddles so sexing is a bit difficult

You may want to post on the serama thread as another option. I think as long as the combs are not red, they can be somewhat large at an early age. My friend got 2 serama pullets years ago and I was sure she got stuck with cockerels. They both had large combs but not red. They seemed to mature at an early age, and have gone broody frequently, but with no rooster - they are wasting their time.

I have 5 seramas 5 weeks old. I've known that Cowboy was a roo for a couple of weeks now. He developed a huge comb early and he's already very protective over the other 4. He made his first attempts at crowing today. It was so pathetic! lol...I think you have all hens, from the pics..
