The counters on paraffin cutting machines really drive in how many cuts you've made, but I'd do 1000 paraffin sections over 200 cryostat sections! 14 micron thick sections with your hands in -20F for 3 hours sucks. Especially when your tisssue slices fall off during fixation and you have to do it all again!!!! Ugh, heaven forbid you used the last block and have to start new mice breedings to get more tissue. Plug check 9am daily and I smell like mouse urine all day.
I so understand no bands on a gel too.
Do have to say the outfits are hilarious, bio hazard bags, table top "potty pads", and her western outfit.
I swear all PI's say first author first year in their lab. What they really mean is 1st author the "first year" that you are "in" their lab "after" ALL coursework is done. In other words. year 3....

I can relate to almost every scene, including the what's in the box, what's that smell, and especially bad project.... we have insane mice that do flips too.
Seems like they have a whole ton more fun in their lab though, and I thought I had a ton of sticky notes.
