We have a 4 month old EE rooster that’s very aggressive to all the birds but particularly one bird, a fellow EE. She’s all white so we think he can see her better. He pretty much chases her all day and she will ride in the box to get away. He’ll also hide in the box almost like ambushing her. Will he calm down since I know he’s just getting fertile? Would separating help or hurt the situation? He doesn’t attack me and I go in and hold him almost daily and he doesn’t mind. They scream so loud, is it just scaring them or actually hurting them?

He's being an idiot adolescent, with no adults to help him develop better social skills. Separate him, not your pullet, and see if some time away will help. And, having lots of space, out of sight areas, and multiple feeders and waterers all will help.
Is she being injured? If yes, I'd wonder if he's worth keeping. As it is, see if a time out for a couple of weeks helps, or move him on.

Yea I was never going to move the hen. She’s not hurt but just being terrorized so much. I’m afraid separating might make him worse. This was one of my last hopes seeing if someone could help or he’s going on Facebook marketplace

Did you try this already or do you just suspect him to do so?

But if you do already have a supposedly nice rooster (otherwise you would not still have him), what do you need the other for anyway? There's a lot of very nice recipes to choose from: https://www.backyardchickens.com/forums/egg-chicken-other-favorite-recipes.25/
No we have seen him attack the adult hens too but he will not come close to the other roo. We wanted to keep him mainly to make babies and He’s a Americana so we wanted to have part Americana babies.
