Do you connect the wood frame to the cinder blocks on the Run?

Did y'all connect the 2x4's to the blocks supporting the run? If so, how and with what?

I didn't.

The weight of everything holds it down - and the hardware cloth goes down in to the ground 12-18" and wraps around the whole thing which helps hold it together. (Need to make sure the frame lines up with the outside edge of the cinder blocks so you can get the hardware cloth tight.)

Ditto on what FridayYet posted. My coop is 10x8 with 2x6 floor joists, 1" flooring, 2x4 framing, 1" board sheathing, 5/8" plywood roof and asphalt shingles on top. No need to anchor it. The weight will hold it in place.
