How much square footage of house/run do you need to keep 12 chickens?

Four square feet per standard size chicken is recommended if they have access to an outside run. So, you'd need 48" square feet.

Coop - 4 sq feet per bird
Run - 10 sq feet per bird

Bigger is SO much better - cleaner, healthier, easier for you to move in their space.

Our coop is 48sq' - so theoretically, can hold 12 birds. Yeah, 11 pullets (3mo) were crowded. Way too crowded for this rainy week we've had. Coop's too small.
Our run is 80sq' - so theoretically, can hold 8 birds. Yeah, 11 pullets (3mo) were getting crowded - run was way too poopy and they ate all the greens too fast. Run's too small.

I'd say double those figures above.
