I have a a window on the North side, one on the East side, and I cut an 8x12 inch vent on the South side, about 6 feet up. The entire shed is 16ft 2 x 8ft 2. Their enclosure is 8ft 2 x 6ft 2. It’s enclosed by chicken wire(the shed is sealed up tight) and all the windows/vents are on their side (east) I could immediately tell a difference in temp when I cut the vent yesterday(covered it with 1/4 inch hardware cloth). Do I need more vents?

Your ventilation sounds good to me! I personally have a cross breeze in my coop (one vent on the North and South side). If you can notice a difference in temperature, your chickens probably can as well.

The more the better IMO....good to have them weather proofed against rain coming in and be able to close most of them off in winter.
Posting some pics of your coop inside and out will garner suggestions of waht you might be able to do.
