What killed my hens??

I live in southern VA. I have 12 full-grown hens and 1 rooster whom I used to lock up at night but allowed to free range during the day. We live in the country but so far there hadn't been a real daytime predater attack, until last month. In one day, I lost two laying hens. The others seemed a little skittish but didn't seem to care beside that. I searched everywhere but couldn't find any feathers or sign of a fight. Naturally, all the chickens went on lockdown and haven't left their yard since. I can tell they miss the outdoors beyond their enclosure (which isn't very big but I was planning on building a larger one) but I've been skeptical about letting them out of the run. I think it was an owl but I really am in the dark about this. Oh, and there are a few owls in the woods (there has to be two of them) that won't stop hooting. Does that pose any threat? I'm not sure if they attack during the day or what but they sure hoot during the day. Anything you guys know would be great. Thanks. Sad about my beloved hens :'(

Thanks for your advice. The hens are safe in their coop and I don't think I'll let them out for a while.
