A couple of days ago I put the six new, young pullets in with the flock after keeping them separated by chicken wire for a while. My older flock numbers ten, one is a rooster. Aside from a few pecks/small amount of chasing {plenty of hiding spaces, ways to get away} it's going really well EXCEPT for one hen who is really being an arse to them when she can. She's second lowest in the pecking order, my personal favorite too sigh.

I'm just wondering if this should keep up in the next day or two if I should put her in the house for 24hrs or so and then put her back in hopes of it knocking her down a few pegs? Or should I just let things be? No blood has been drawn, no injuries... Miss Penny is just being a jerk to them.

As long as there's no blood, I wouldn't worry to much about it. As you said, she's 2nd lowest in the pecking order and wants to retain her hard won status!
