5-6 weeks old at least, closer 8 weeks is easier because the vent is more developed.

You either have to vent sex them, listen for sex specific vocalizations (the roo's crow, and the hens LOUD cricket chirp), look for roo's mounting hens etc. When vent sexing you should always double check yourself because hens that have been bred recently will have foam in their vents, just like a roo would.
