I have 1 Rooster. I built him a box fully insulated. I have 2 floor vent registers on the upper part of 2 walls. Question is will he he warm enough inside by himself with no heat source?

Is he your only bird?

What are your worst winter conditions? Wind? Rain? Snow? Temps?
If you get high wind and wet will it be able to blow in the vents? If yes then you may need to put some sort of baffle on them to slow the wind and block the wet. A lot of people use the blue "cut to size" furnace filter material.

I have 7 hens. I split the end of my run which is wrapped in plastic with ventilation around the top. I split off the end of the run for him to have his own coop and run area. I also have those clear plastic magnetic air vent diverting things to helpfully help push too early crowing downward. I like the air vent idea never heard of it. Thanks for responding.

Also I am in N.J. we get in the teens alot at night. Sometimes the real feel could be zero but not often.
 
Is he segregated because neighbors would complain? I understand some people have that need to separate a rooster. With your temperatures I think he will be okay just be sure to block the wind from coming in those vents.

I'm just trying to be a good neighbor and reduce the sound of early morning crows. I only have 7 hens and don't want them over abused. Also I do not have the desire to have fertilized eggs. It was 15 degrees last night when I let him out this morning his outter feathers were a bit cold but I put my fingers down through the feathers to skin and he was nice and warm. Thanks again for your advise and time.
