Have you picked her up and examined that more closely?
Could be a piece of debris or a small scab.
Photo is not that clear, but it looks like you may be able to just wipe it off.

To me it looks like a small scab. Maybe a fly bit her there or she got pecked by a fellow chicken. Doesn't look like anything to worry about
