frostbite and ventilation

The little triangle vent is insufficient for the number of birds you have. Without measurements I'm not sure exactly how much you have, but it's probably closer to sq inches of ventilation, when you need sq feet of ventilation. For your flock you want roughly around 6 sq ft of draft free ventilation open 24/7 to allow moisture to escape.

If feasible I'd move the waterer out as well, as that could be contributing to the moisture content inside the coop.

As far as the frostbite, best to not touch it. The tissue is already damaged so rubbing at it will only do more damage. If parts of the comb tissue die, the damaged areas should drop off on their own.
 
The waterer might be your problem source!.
I keep my heated water separated from coop by a plastic curtain.( Then I pump to coop in a closed PVC system) . When we had horribly cold temp last year -23f only the water side had frost on walls.

My girls have had it get to 17f in their coop with frigid wind chills outside and no problems in 2 winters
