One of my hens has recently been showing a pale, shrunken comb. She is also losing weight, and I heard her cough once. None of the other hens seem to be affected so far.

Coughing once is not significant, but if she is coughing or sneezing more often, it could be a respiratory infection. Since she is losing weight, I would examine her crop to feel if it is empty, full, soft, or firm. A crop or gizzard blockage can cause them to starve. Separate her and try to get her to take water with electrolytes (SaveAChick) or just water until you have some. Offer some chopped up egg, and try putting a lot of water in some chicken feed.) Look her skin over around the vent and elsewhere for lice or mites or little clumps of white at the base of her feathers. Look for dirty nostrils or nasal drainage, and watery eyes. How old is she, when did she last molt, and has she laid eggs recently?
