I got 3 new pullets yesterday, all Easter Eggers, all lived in the same coop. One is now sitting on the roost with her eyes closed, and every time she takes a breath she opens her beak. She's very lethargic and not really alert. I don't know what's wrong! She was totally fine yesterday, her sisters are fine, and I found no abnormalities when I inspected them. She doesn't have diarrhea or bloody stools....

I'm sorry to hear that.

Probably, the hen was getting sick and when you moved her to a new coop it stressed her and her immune system dropped, making her get sick faster. This is an emergency. Breathing problems can be caused by:
Over-heating
Respiratory infection
Gapeworms
Fowl Pox

If she has an infection, I would get her onto antibiotics as fast as possible, as birds have sensitive lungs and can die quickly from infections.

I hope she gets better soon. Best of luck!

What kind of antibiotics? Now she's off of her roost and drinking water, but eyes are mostly closed/watery/has a runny nose. Do you think it's most likely a respiratory infection?

What happens once her symptoms go away? I know she'll be a carrier of whatever infection this is, so do I still introduce her to my flock of 5? What about the other two new pullets? Do I treat them although they have no symptoms? After 30 days of no signs of infection can I introduce those two into my flock or are they going to infect everyone else? Lastly, is it best to cull those 3 for the sake of the rest and get new pullets? I'm worried one may kill them all.,,,

Some causes behind respiratory infection can spread very easily, other forms are just caused by stress and moist or dirty conditions. Continue keeping them separate for the 30 days, if they show no symptoms, I would consider it safe to introduce them. You could try getting them tested.
