I have one roo and like 40 hens and they lay good for the few 5 or so that do lay because most of the other hens are older and retired and some are young and new and aren't laying yet so really i don't have that much advice

Well it's a barn so idk 50x50? What does this have to do with the hens to rooster ratio?

If it's actually 50x50' should be plenty of space,
and to make separate enclosure if needed.
Space can have an effect on multiple males getting along.
