Is this frostbite?

Looks like it to me. It may turn black .... or if not too damaged ... recover. You can Google chicken comb frostbite and click on images. I've had some of my chook combs get a bit frostbite damage but they did recover without much trouble.
 
I think I have the ventalation covered better than average. Alas, winter in Wyoming, single digit highs, and sub-zero nights

there are 2 (one oscured) of the upper vents and the window on the side is hardware cloth. The other two windows have plexiglass pannels in place for winter

What is your humidity like there?

Usually lack of ventilation is the culprit, but seems you have that covered!
