My buff orp hen sounds like a duck. Is she sick?

If could be a respiratory problem of she could have something stuck in her throat. If it continues more than a day or so I would give a listen to her breathing to see if you hear and wheezing or crackling which would signify an infection of some sort. A vet would be needed for treatment.
 
I just picked up my hen and she also had clear fluid leaking from her vent. Should I separate her from the rest of the flock? Is this serious?

When was the last time she laid an egg?

With your hot weather, I would be careful examining her during the hottest part of the day, you don't want to stress her.
Check the crop first thing in the morning to see if it has emptied, feel the abdomen for any bloat/swelling. The clear liquid from the vent - was it like egg whites or just discharge, was it time for her to lay and egg or just after when you noticed it.

Sounding like a duck, well...it would be good if we could hear that. I have one that is 3yrs old and has always sounded like a goose at times (mostly when she sees me:) ). I have another that plays broody and can quack/honk pretty good instead of clucking. Can you post a video of her? (upload videos to youtube, then provide us a link)
 
My Buffs make a honk-like noise, but only when it's really hot outside and they've been panting. I often hear it coming from the nestbox because it's hotter in the coop. And I saw one actually throw up water from overdrinking. Hopefully, this is the issue with yours, because it can be remedied easier than a really sick chicken.
