I'm new with all this.

I just build a new coop. It 4' by 6' Is that big enough for 10 chicken?

The nest box is on the outside. I been doing some reading. I like the DLM how deep are we talking about? 2" to 3" I'm going to be using saw dust. I did make some mistake

on the nest box but I'm going to try it first. I put the nest box level with the floor.

Yes that's plenty big enough, your nest boxes should be fine... others do it to, I believe DLM suggests 4-6"

If you go by the standard, you should have approx 4 sq ft per bird, INSIDE the coop. You can have some lee-way on that measurement, but overcrowding causes pecking & fighting due to cramped conditions. The coop you built will comfortably house SIX birds. Like I said, though, just keep an eye on them for any injuries/bleeding with 10 in there.
How big is their run?

If they are only ever in the coop to lay eggs and roost at night and have a large run and/or free range during the day, you can probably get by. But most likely, no it will not be big enough for 10 chickens. If they have to spend any amount of time inside "cooped up" (such as severe weather, snow, et al) then no, it is definitely too small. My coop is 4x5 with a 10x10' run and has 7 chickens in it. It's not big enough and we are seeing some problems with one hen plucking and barbering feathers from all the others (I know it's her only because she's the only one without damaged feathers). With nice weather coming up and the chickens spending a lot more time loose in the yard I'm hoping it won't be as bad. But I'm still planning on reducing the number to 5 by this winter.

My run is about 6' x 12'

My coop is 7x7 and run is a little over 12x16 for 10 birds

But, looking at this we all assumed you were asking about standards... are you?!?!
