Will this hold 2x4 welded fencing on 2x4 wood?
If not, will screws be good enough?

I don't think swingline type staples are strong enough to hold a woven wire fence. You need fencing staples. You can use a hammer on them but a 2X4 isn't likely to be stiff enough to drive a fencing staple into so you may need a staple gun.

Screws don't work on wire unless you attach another strip of wood over the wire with the screws.
Meant to say welded wire, not sure if that makes a difference
 
Staple gun Staples won't hold fencing. As noted already, fencing staples that hammer in. You can use fender washers with screws. They're the larger body washers with small hole in center. If you go that route make sure you get galvanized or coated screws for outdoor use.

Standard washer on top, fender on bottom, for reference.
3/16" hole in washer works best. If using coated deck screws you can use 1/4" anything larger and the screw will pull through and not hold.

It's a good way of anchoring the ends. Truthfully the staples are cheaper and hold quite adequately. I use the screw and washer combo sparingly, usually on hardware cloth. If you do use it. Drive the screw into a corner of the mesh so the washer covers a vertical and horizontal wire.
 
Agreed. The screw and washer method is much more expensive than fencing staples and not very effective on welded wire. It works quite well on hardware cloth though.
