how long do I need to keep a hen seperated from a rooster and then put her with another rooster to make sure that she is bred to the latter rooster.

Sperm usually lives about 10 days in a hen. Of course, there are cases when it has lasted 3 weeks. Generally, if a hen is mated by a second rooster, he is usually the sire of the chick (freshest sperm usually wins the race). This is not always the case, but most often it is. There is no absolute.
