I have 8 Rhode Island Red chicks about 3 months old. what is the best way of telling a rooster and hen at that age?

At three months your young roosters will have larger, redder combs and wattles. Their legs will be thicker, they'll be more upright and may be starting some saddle or hackle feathers, but they're at the low age for those.

How do you tell when there first born?

Unless they are specifically created Sex-links, you can't. Being trained in vent sexing, that is only 90% accurate.
