Might help if the girls can get away from him, something to break the run up and his line of sight, bale of hay etc.
 
I'd start like you said, get that roo out of there. Long enough for the girls to heal up completely. I don't know about the peepers but if I had a roo that did that, I'd be looking at freezer camp.

NO sorry but it does not sound like your roo is being good with the girls at all. Something changed him into a very bad roo. There are a lot of good roos out there so if the peeper trick does not work you are better off eating him before he eats your hens.
 
There's no specific reason why he's going for the vent. Maybe he has a thing for fluffy feathers? I've dealt with feather picking all throughout my owning poultry for one reason or another (I think I'm just unlucky) and have seen it start anywhere from the back to the neck to the FEET. Never had the vent though.

It just starts where the pecker want to start. No real reason. He just decided he wanted to peck feathers from the vent. It very easily could have started on any other area of the body. It's good that you're keeping him separate. Best take him away before he can inflict any more damage on your birds.
 
High production birds are more prone to cannibalism. The research I did suggest that when you increase the production of eggs in a breed you can also increase the bird drives to be territorial. Often picking at the vent areas can occur. For example production reds are known to do this. My leghorns started it at 2-3 weeks old. It is just what they do when kept in larger numbers and that is why they are de-beaked. I have the peepers on my 22 hens and just took them off my rooster who also picked so far he has not repeated the behavior after wearing them for 3 weeks. All the hens have also healed.
