Yes, they do.

Some will tell you they won't, but when the "mother ship" tells that hens it's time to be broody, she's broody, and that's that, roo or no roo.
 
So, basically if there is a roo, there's no way to know whether she's sitting on fertilized eggs unless you've been keeping tabs on who he'd been doing the BD with?

If i'm seeing all fertile eggs, then i assume that the eggs my hen is sitting on are fertile.

You can also candle them after 10 days or so and see if they are developing.
