Can you build your run on a small hill?

Yep just make sure there are no gaps under the fence

Most of my chicken yard is on the side of a hill. Not a problem for the chickens. In the steeper areas everything tends to move downhill with chickens scratching. It can pile up against fences if you don't plan for it.
Also chickens can fly really high especially if they have a hill to fly from. Just something to consider if the run is not covered and fences not too high.

Can be a good thing to prevent flooding/standing water......but may depend on what you have planned and the contour of the 'hill'.
Pics of area and coop plans would help immensely to offer a viable answer to your question.

The worst place you can put a run or coop is a low spot where water stands. A hill where water drains away can be a great location. As Aart said there may be other issues, mainly concerning erosion, depending on how steep that hill is. There are techniques to handle that, which to use depends on how steep it is.

Chickens scratch and gravity moves things downhill. You may need a barrier along the bottom to catch that stuff. If it is really steep you may need terracing. If rainwater runoff can go through it you may need a berm or swale above it to divert the water.

To me the possibilities of a coop/run on a hill are great but the devil is in the details.

Mine is on a hill. The hardest part about it is that there are dips and such that you have to take into account when predator proofing, so sometimes you have to be a bit creative with trying to ensure your coop is level and your run does not have gaps that critters can use for entry.
