My chicken coop is upcycled from my kids’ old play set. Therefore, the floor is the ground. Can I use the deep litter method in this instance? Also, what if I don’t need fertilizer for my garden, what should I do with the litter that I remove?

If your city/county does composting or lawn litter pickup you can toss it in there, or if you have the land for it, just dump it somewhere in the field and let the weeds enjoy it. Or if you have friends/family that garden I'm sure some of them would be happy to help clean for some compost!

I don't garden & rarely remove any "litter", maybe once a year ... I've used to fill in low areas in my yard, used as mulch around my plants and as rosemarythyme said pile it in a section of your property. DLM works great for me & keeps cleaning very easy.

Wonders if there would be problem if it's got chicken poop in it?

You could do deep litter in your run too, and just add it to that.
