Help!!

Anything that gets frostbite will die and fall off, even toes and feet.
Affect her? I’m not sure. I thought roosters fertility could be affected by a frostbite comb but this came from an old farmer and these thoughts aren’t always correct.
I know you have ventilation but are the drafts blocked?
What drafts, I just keep the door open like this during the night and take the water out
 
I don’t keep water in the coop and I keep the door open since I have two doors for the coop. The other door I keep open is surrounded by a run THat is predator proof. Also they do roam outside. Is there anything I can do
But will it grow back or that will be permanent? Will it affect her though? Because I don’t know how to fix it
I would leave the combs alone.
You will want to try to keep them out of direct drafts at night. You are in Canada so your temps are likely quite cold. Frostbite can be common. Some breeds that have pea combs or are designed for colder climates like yours (Chantecler) have less problems with Frostbite, but it can still happen.

Once tips are lost they will not grow back. It should not affect her.
 
With the door open, is there a draft coming through that door? Drafts are not good for chickens and can cause frostbite without water in the coop.

So will my chickens whole comb fall off.. and when will it fall
Just the parts that die. Whatever is the normal red color will remain unless it gets frost bit as well. It will fall off when it's completely dead and there's no more blood flow. Could be a few days, could take a few weeks. Usually you will just notice suddenly that it's no longer there
