Hen keeps one eye closed

I tried to gently brush it out without success. I am not sure about the nictitating membrane (I did not know what that was when you first mentioned it). Each morning I flush her eye with warm water, hoping to dislodge that layer. I don’t think it is pus but that is based on experience with infections in humans, not chicken eyes. LOL.
Just a quick note, not sure if you know or not, but chicken pus will be somewhat solid, like rubber or cheese. It is kind of shocking when removing a piece of chicken pus because it isn’t what you expect. It can be very tough to get out due to the consistency. That’s why we keep suggesting the warm compress to try to make it a bit softer and perhaps easier to get out.

Follow eggcessive’s advice about saline and keep trying to work the material out. Seems like you are making progress with it. Keep at it with the ointment, too.
