Oh yes.. if the hen wants to lay in THAT nest.. she will, even while the broody is still on it. It's a very common occurrence actually, and like others mentioned, it's practically a necessity to at least mark the eggs set for hatching for this reason.
However this is also undesirable(other hens having access to nest) for another reason- if hens try to lay in the same nest, and they do so while the broody is still on, it can and often leads to broken eggs. Quite a mess.. These are the common reasons people either isolate the broody hen/her nest from rest of the birds or simply move them to someplace by themselves.
