That won't be enough.

The coop is 10x12 feet on four sides, not on one side. How tall is the coop?

If it is 10 feet wide, 12 feet long, and 8 feet tall, then you have 80 plus 80 plus 96 plus 96. One coat. Probably needs two. I'd get two gallons.
 
If you're painting the floor, yes. However, if you intend to paint the walls, by my math, you're cutting it really close. Our coop is 5' x 8', and we just BARELY had enough to paint it with one gallon. Next time I'm getting two and keeping the leftovers for touch ups.

Figure out the square footage for each wall, then add them up. If one wall is 8' high and 10' long, that's 80 square foot. Once you have each wall add them up. No need to do anything fancy like subtract for windows and doors. You'll just have a smidgen more paint left.
