I set my eggs on the 20th of March. But I forgot to let them settle pointy end down for 12 hours. Instead I let the quail eggs settle pointy end on top for 12 hours. Would they not hatch cause of this?

No, this alone will not make them not hatch.
 
The chicken eggs were from us. We did not even let them settle at all.
I don't let shipped quail eggs settle. They go straight into the incubator as soon as they arrive.
I have great hatch rates.
 
Then why does it benefit the eggs?
Letting the eggs sit with the pointy end up benefits the egg in a zero ways.

I'm not sure I'm understanding what you're asking.
