we have found one of our hens has lice. We have treated her and the other chickens (we have 5 hens plus a rooster), but she is the only one showing any signs of the bugs. Is it possible she is the only one with the infestation?

Yes, it's possible she could be the only bird infested. You did the right thing by treating all your birds. Retreat all of them again in 7-10 days. Do the same thing inside their coop, nests, roosts and repeat again in 7-10 days.
