If you take a warm wet napkin to the spot does it come off at all, or color the towel red, like dried blood?
The lesions or warts eventually dry and fall off onto the ground and are very infective. If they are eaten by a chicken or turkey, then it develops into wet pox. Wet pox is worse than dry pox and is life threatening.

I was just wondering if it actually is pox. Like, maybe it is just a scab from a peck wound or something. But yes, if it is pox I really hope it's dry and not wet. :fl
 
I've dealt with fowl pox in chickens and it passes slowly through a flock. Hens are too grumpy to lay eggs, are skittish and look terrible. Iodine can be put on the lesions to help dry them up quicker, just dont get it in their eyes. Black shoe polish can be used in the same manner, it also prevents picking and pecking the infective scabs by other birds.
 
I found this thread of a person who was afraid their turkey had blackhead disease. (Has spots all over its head). Turned out to be fowl pox. Does the spot your turkey has look like the spots in the picture the OP posted?

Thank you all for the info! This has been very helpful to consider other options it could be. Our turkey’s spot seems darker than the ones in that picture. I haven’t tried dabbing it with a cloth or napkin, that is a good idea - though I doubt she’ll let me close enough right now! We will keep monitoring it to see if it spreads or grows worse. So far she hasn’t shown any other symptoms of being ill. Here’s hoping it’s just a scab and nothing more serious! Although as far as being pecked, she’s currently the only turkey amongst a flock of ducks.

Is there anything in her home she could have poked herself on? I'm don't think ducks can "peck" but I've never owned any so I'm not sure. I know they have toenails. Could one of them have flown up and poked her maybe? Idk. Seems unlikely.

She free ranges around our property so she could have injured it on anything.
 
Update: we just spotted a black spot on the wing of one of the poults - apologies the picture is terrible, the mama was taking them into the brush when we saw it so we didn't get a great view. Also here is today's photo of the black mark on the female :(
