I was making an egg sandwich and I saw this, is this a sign my rooster is finally mating with my hens or just a random blood spot?

I agree. That is a meat spot, nothing to do with a rooster. It is safe to eat but I'd pick it off before I ate it just because I figure it is gross.

You do not need a microscope to tell if an egg is fertile, you look for the bull's eye. This thread explains it and has photos.

When you crack an egg to look for the bull's eye it may be on the bottom. I gently turn them with a spoon to find the blastoderm or bull's eye.

The poll question is, Is this egg fertilized? There is no way to answer that based on the evidence. What we are seeing is a meat spot, which clearly has nothing to do with fertilization. @Ridgerunner is telling the OP what to look for in a fertilized vs unfertilized egg, in order to answer the question as asked.
