I usually wait until daytime, when they sleep.

I cover the trap with a blanket when I find the occupant (not a blanket to use again maybe)....

And then gently open one door and back away.

I've done this a few times and thank goodness the skunk still is calm or sleeping and leaves the trap when it wakes up.
 
If you move slowly and don't startle the skunk it probably won't spray. They only spray if they are very angry or very frightened. I saw a sement on Mythbusters once when they wanted to try out remedies for getting rid of skunk smell. So they needed the skunks to spray. Had a devil of a time. BTW, if the worst happens and you do get squirted, the only thing to do with the tomato juice is to make a Bloody Mary out of it. It is useless for skunk spray. I can't understand these people who when they see a predator, the first reaction is to kill it. Fix the chicken pen so they can't get in, for heaven's sake. Killing them just lets another move in and you are no better off than when you started. BTW, we have lots of skunks, coyotes, hawks, and possums. Haven't seen any coons but we probably have them too. None of these critters has caused us any trouble at all.
 
Shoot it in the lungs, NOT the spine or head, damage the nervous system and it'll spray.

For yall who disagree with shootin 'em... I've already been chased by 4 rabid skunks this year, if folks would kill the pests they caught instead of lettin 'em go, there would be a lot less of that. I've had rabies shots, they hurt like the devil, and I aint to keen on goin back for more. But if yall want the same, go for it, just make sure and dump 'em on your own land so you, your kids and your livestock will get bit and not everyone else, then see if yall will release after that.

I have never seen a skunk around here ... DH has but I haven't. Thought we were talkin' cats lol
 
Caught four of these "kitties" this spring myself. One was an albino. (PIC attached) Beautiful animal.
Used 3 pole extension tool which I use to change out my high out of reach light bulbs with along with 2 length pole extension tree trimmer. Pushed in the spring loaded latch and pried door up enough for them to escape. Might want to let them settle down a day or so before approaching. If they start stopping their front feet and turn around on you back away. They have very poor long distance eye sight.
Use an old shower curtain or pig piece of plastic if you wish to walk up to the cage to cover up or to release your kitty. Don't let them see you.
Got sprayed at ..not on once when I turned the cage over in the process of opening the latch.
