The poison will not kill the eggs. OP needs to wait 7 days and treat again to kill the mites that hatch. I would treat again a third time in another 7 days.The spray should kill the bugs on contact.
It's the million eggs that are hatching daily that you have to knock down.
I would say you should notice a change soon.

Assumes that they are red mites. northern fowl mites will hide in the vent.See the last post.
They live in the coop and only feed on the bird for so long each day.
