If the shot is placed correctly, the skunk will not spray. What will happen is that all of the sphincter muscles will relax with death allowing the scent glands to discharge but it will not be as a spray.
 
If the skunk has sprayed once, it takes awhile for them to make the spray again. They don't like to spray, but will if they need to. There are lots of warning signs they will give you first. Backing up, raising fur, even standing on their front feet doing a head stand! But if that butt aims at you, back up! And you should not relocate it. It is illegal in many states and a bad idea. It will disrupt the wildlife of the area you relocate it to. Other animals will have to fight it to show their boundaries. Shoot it but good luck!
 
I saw my dog playing with my little black cat and then felt my little black cat rubbing my leg. Yep Pepe LePew. Vet said use Dawn dishwashing liquid as fast as possible as skunk spray is oil based. It worked pretty well and she only smelled vaguely skunky for a few days. Moral to the story, buy Dawn when you get the trap. Oh and don't play with black cats.
