I killed all the spiders by whacking them with sticks and removing the eggs sacs although some still remain. I'm thinking about using spider killer spray and spraying it all over the top of the inside of the nesting box as that's where I saw them all. Im going to let the spider killer spray dry for a couple minutes before allowing any hens or roos into the nesting box. Is this OK to do by using spider killer spray in the nesting box?

I absolutely would not use any bug sprays. Chickens love spiders by the way, unless you've IDed them as poisonous (like black widows) I would just knock the spiders over to where the chickens are and let them have at.
