Just the membrane. Let it dry then cut out an area at least an inch square were there had been blood. Put that in a small zip lock bag and attach it to the lab form for that emu. Leg band them or do something to identify them. I just used the "Egg B" name. Go to Zoogen's site here:

Choose either a worddoc or PDF form. Print it out and fill it in. Staple the sample for each emu to their own requisition (I also wrote "Egg B"
on the ziplock bag) Send it in. I was anxious so I sent it in an overnight flat rate envelope. It goes to a post office box in Davis, CA. The lab owner picks up the mail on her way in each morning. You should have results 24 hours after they receive the sample. Write your phone number down in big letters if you want them to call or write down your email address and ask to be notified that way. A week or so after you get the initial report you will get a nice certificate in the mail. That should be signed over to the new owner should you ever decided to sell them.
Keep what membrane you have left in case they need more sample. That only happened to me on one's I feather sexed.
