Methinks you have a bigger problem than the chickens. I would rehome the chickens at least for a little while, but it sounds like your neighbor has, er, mental issues. When the reaction is so far out of proportion to the "annoyance" there is something else going on. I would distance myself as much as possible and keep my eyes open. Once the chickens are gone there may well be something else she is going to fuss about. It doesn't hurt to keep a journal either.
 
If you didn't look up your ordinances personally, and are just going on what you have been told by others, you definitely should do your own research. My husband was telling me all these "ordinances" that he'd heard about from people in the area, most of which are not true...check it out to be positive, because frankly- one acre is plenty of space for chickens.

I hate crabby neighbors and I have to say you handled this very calmly - kudos to you! Wouldn't it be something to get the town to reconsider their ords and then get enough roosters to peeve her off - legally?

As my father (God love him) would say: I'd make, taking down that woman, my habit. I'd get those laws changed even if it killed me.

If you're not feeling as fiesty as I appear to be... then I'd go with the other opinions... rehome the roo, send the girls on a temporary vacation until everything blows over and then bring them back when that crochety old fuddy duddy gets over herself.

Look on the bright side......You won't have to house sit for her anymore.
Also I would do as someone else said and check the ordinances for yourself.
Do not go by what someone says to be true.
