Sounds like fox to me. I have lost that many in a matter of days to fox. I hope you figure it out.

They're in a four wall barn/coop, doors locked & shut tight every night.​

I am glad to see someone else mentioned fox. That is what I would suspect first. They can even catch guinea. They are so sly they took 30 of our layers before we even new it. They were bold enough to try to steal chickens I was watching.

If the barn's locked, it must be something going over or under.

I hope you catch what ever it is and then reinforce the area it's getting in.

That's a whole lot of birds to spend time and money raising only to lose to a preditor.
