Maybe try an antibiotic as a last resort. And in case it does not improve his condition in a timely manner, I would help him out of his misery.

There are several quick methods to choose from without having to spend on another vet visit.
 
Sorry to hear about your rooster. Did the vet seem knowledgeable about prolapses and chickens? I had wondered about a tumor, but the suggestion of a prolapse that had become necrotic seemed more likely. Cancer of different kinds is really common in chickens. I would have to agree that putting him down might be the kindest way to keep him from suffering. It is never easy, but sometimes we have all had to do it.
