If your hen has had those spots on the comb for several months it might be fowl pox. Its a fungal infection when a cut or wound is exposed to bacteria from your hens feces or just anything dirty. The treatment takes a while and there is no known cure. But I advise you apply antiseptic solutions for hens on the wounds on her comb. Also disinfect her habitat and keep things clean. Lastly, I would supplement her diet with extra protein, maybe a hard boiled egg could work. I also like to treat sick hens by giving them the "Save-A-Chick" electrolyte powder in their water, it really helps. Good luck, I hope your hen heals nicely.
If you apply Blu-kote to the comb, it's both an antiseptic and an anti-fungal. That way, if it's pox, it will help dry out the lesions and if it's a fungus (favus), it will kill it.
If you don't have Blu-kote (Blue Lotion) on hand, try vaginal yeast cream or athlete's foot cream if the antibiotic ointment doesn't improve it.
It looks like a very mild case of favus. Miconazole,or clotrimazole cream can help if applied with a glove daily. You cannfind one of those even at Dollar Tree.
