Is this a chicken ear infection?

This morning I had noticed my rooster ear with this yellow thing I’m not sure if it is an infection?

Yes, it is. I would work on loosening up the gunk so you can pull it out. Then I'd dilute down some hydrogen peroxide and rinse his ear with it. Giving him some plain greek yogurt to may also help fight off infection.

Remove the gunk, use hydrogen peroxide, undiluted to cleanse once, then apply Triple Antibiotic Ointment into the ear.

I just pull it out gently. If it needs to be loosened, apply a wet paper towel to it.

I would drip some warm water to loosen it up and then work on slowly pulling it out.
